Isoelectric
focusing is used for amphoteric molecules (having
positive or negative charge depending on the pH) such as proteins,
enzymes and amino acids. The
Scie-Plas 2D Electrophoresis system comprises everything needed to
run 2D electrophoresis. Both tube gels (first dimension) and
vertical slabs (second dimension) can be run using these units. With the Modules available, 2 slab gels, or 10 capillary gels can be run in the tank at any one time. The modular and 2-D systems are available in two sizes 10 x 10 cm and 20 x 20 cm to complement the TV series Vertical gel electrophoresis systems. |

| First Dimension - Capillary Tube Gel Module | |||

The Scie-Plas Capillary Gel Module which performs the first phase of 2-D electrophoresis (Isoelectric Focusing - IEF), allows 10 capillary gels to be run at any one time, using either the 10 x 10cm or 20 x 20cm TV systems. Essentially these gels differ in the length of the capillary tubes, which are 80 or 170 mm long, allowing a high degree of versatility in the separation desired. The capillary tubes have a wide capillary bore diameter, which greatly eases the extraction of the capillary gel once the first dimension - IEF phase is completed. After focusing, which typically takes between 3 to 4 hours, the tube gel can then be extracted and the samples separated on a slab gel for the second dimension, 2-D, size determining phase. The second separation can take as little as 1 hour to complete and therefore a complete 2-D separation can take as little as 4 hours. | ||

The 10 x 10 cm and 20 x 20 cm vertical electrophoresis unit can run two gels or commercially available cassettes for the separation of nucleic acids or proteins. The running tank of this unit accepts the Capillary gel module and the tube gel module. The running buffer can be pre-chilled prior to electrophoresis or additional cooling can be achieved by using a bottom cooling block, The super-cooled versions are set up for use with a chiller circuite. Gels can be poured directly into the gel running frame which connects them to the top buffer tank. A leak proof seal is obtained with sprung clamping bars and screws. The complete gel assembly is then placed in the bottom buffer tank and can be fully submerged with the running buffer. | ||
